Flexi Says: The role of primase enzyme in replication is to add RNA primer on the lagging strand. If the enzyme primase is removed from the cell, it would not allow the initiation of replication of lagging strand but the presence of primers will let them to anneal to the corresponding segment of DNA. DNA polymerase is then able to use the free 3'-OH group on the RNA primers to make DNA. The process is possible only in vitro.
